Objective:
To describe the procedure to be followed for cleaning, operation & monitoring of laminar air flow.
Scope:
Applicable to Laminar Air flow unit in Microbiology Laboratory of Quality Control Department.
Responsibility:
1. Lab Attendant : Cleaning outer surface of LAF daily .
2. Microbiologist : To perform the analysis and to clean the bench of LAF before and after completion of work.

Procedure:
Cleaning:
1. Daily
2. Clean the outer surface of the laminar airflow unit (LAF) with cotton cloth soaked in70% v/v IPA (0.45 m filtered) followed by wiping with dry cotton cloth.
3. Before
starting & after completion of any work, clean the bench of LAF with cotton
cloth soaked in 70% IPA (0.45 m filtered).
Operation:
1. Ensure
the cleanliness of laminar airflow bench.
2. Check
the manometer scale before starting LAF, the scale must show the level as “0”.
3. Switch
ON the main power supply. Switch ON the laminar airflow unit by operating
ON/OFF switch located at upper panel of laminar airflow.
4. Start
the laminar airflow approx. 15 minutes prior to use.
5. Ensure
that the manometer scale of all LAF is more than 10 mm. If manometer reading
falls below 10mm, then inform engineering department.
6. Note
down the reading as per the SOP/QC/066 Titled “Monitoring of differential
Pressure in Laminar Air Flow”.
7. After completion of work clean the LAF.
8. Switch OFF the laminar airflow & main power
supply.
Monitoring:
1. Ensure
the cleanliness of laminar airflow.
2. Switch
ON the laminar airflow by operating ON/OFF switch located at upper panel of
laminar airflow.
3. Settle
plate Exposure.
Frequency : Twice
a week.
4. Expose
2 SCD agar plates in LAF for 1 hour and then close the lid of the plate as per
SOP/QC/062 Titled” Environmental monitoring of microbiology laboratory”.
5. Collect all exposed plate & Incubate the
plates in inverted position at 30-35°C for 2 days followed by 20°C – 25°C for 3 days to
check for microbial contamination.
6. After
incubation observe the plates and count the number of colonies. Record the
results as per SOP/QC/062 Titled “Environmental monitoring of microbiology
laboratory”.
7. Validation
for DOP test, Velocity and Particle count is to be performed by Approved outside
party (Frequency : Every six months).
8. After the validation put the label.
